High-performance liquid chromatography (HPLC) separates components in a dissolved sample. A purity figure on a peptide COA is usually the area of the main peak relative to other peaks detected under that method — not a guarantee of biological activity or complete chemical identity.
What HPLC can support
- Relative abundance of the main analyte versus observed impurities
- Comparison between lots run under the same method
- A documented chromatogram for the tested batch
What HPLC does not prove by itself
- Exact amino-acid sequence (mass spectrometry is typically used for identity)
- Absence of salts, residual solvents, or water unless those tests are listed
- That every later vial from a different lot matches the same chromatogram
