Understanding HPLC Peptide Purity

High-performance liquid chromatography (HPLC) separates components in a dissolved sample. A purity figure on a peptide COA is usually the area of the main peak relative to other peaks detected under that method — not a guarantee of biological activity or complete chemical identity.

What HPLC can support

  • Relative abundance of the main analyte versus observed impurities
  • Comparison between lots run under the same method
  • A documented chromatogram for the tested batch

What HPLC does not prove by itself

  • Exact amino-acid sequence (mass spectrometry is typically used for identity)
  • Absence of salts, residual solvents, or water unless those tests are listed
  • That every later vial from a different lot matches the same chromatogram
